报告简介:

To understand the mechanisms of human Pancreatic ductal adenocarcinoma (PDAC)tumorigenesis,we have established a novel system to isolate and genetically manipulateprimary acinar cells and ductal cells from normal human pancreatic tissue. Our system is able to separate human acinar cells from ductal cells to study the irproperties and their capacity to generate tumors. This has never been achievedwith the published organoid culture, in which only ductal cells can formorganoids. Taking advantage of our unique system, we will continue to elucidatehow acinar to ductal metaplasia (ADM) facilitates PDAC initiation, and willprovide valuable insights on how pancreatitis and other cellular stresses contribute to PDAC development. Our findings hold promise of identifying targets for PDAC prevention and biomarkers for detection of early stage humanPDAC. Continuing research with our system will allow us to establish a full model of PDAC for evaluating novel therapies. Studies of normal gene functionduring development will inform our understanding of the pathological rolesthese genes play in disease states such as PDAC, one of the most aggressive anddeadly human malignancies.
